In this study, the peptide B1 component with excellent gastric mucosal repair activity was isolated, purified, and identified using Corneum Galli Gigeri Endothelium (CGGE) as the raw material. It is mainly composed of new peptide segments such as DYPELS, LPPLEH, SFYYGK, DDDGVGF, VVLPR, and LPYPR. Among them, except for LPYPR, which has been reported to have activities such as lowering blood pressure and cholesterol, the other 5 peptide segments are newly discovered active peptides with the ability to repair gastric mucosal damage. Additionally, based on the GES-1 cell model, we further elucidated its mechanism of improving gastric mucosal damage. We found that the B1 component can regulate cytokines such as EGF and TGF-α by reducing oxidative stress, enhance the synthesis of HGF, TFF, and VEGF, modulate the protein expression of Cdc42, Rac1, and RhoA, associate with the Rho signaling pathway, promote cell migration and proliferation, and consequently facilitate gastric mucosal wound healing.
本研究以角膜内皮(Corneum Galli Gigeri Endothelium,CGGE)为原料,分离、纯化、鉴定具有优异胃黏膜修复活性的肽B1组分。它主要由DYPELS、LPPLEH、SFYYGK、DDDGVGF、VVLPR和LPYPR等新肽片段组成。其中,除了据报道具有降低血压和胆固醇等活性的LPYPR外,其他5个肽段都是新发现的具有修复胃粘膜损伤能力的活性肽。此外,基于GES-1细胞模型,我们进一步阐明了其改善胃粘膜损伤的机制。我们发现 B1 组分可以通过减少氧化应激来调节 EGF 和 TGF-α 等细胞因子,增强 HGF、TFF 和 VEGF 的合成,调节 Cdc42、Rac1 和 RhoA 的蛋白表达,与 Rho 信号通路结合,促进细胞迁移和增殖,从而促进胃黏膜伤口愈合。

Recently, many researchers have found that various peptides have multiple biological activities, including anti-inflammation, anti-diabetes, anti-cancer, anti-osteoporosis, anti-multiple sclerosis. Nevertheless, the detailed mechanism of polypeptide in improving gastric mucosa injury repair is rarely reported.
最近,许多研究人员发现,各种肽具有多种生物活性,包括抗炎、抗糖尿病、抗癌、抗骨质疏松症、抗多发性硬化症。然而,多肽改善胃黏膜损伤修复的详细机制很少报道。
In this study, we are the first to report that the B1 component isolated from CGGE has a reparative effect on gastric mucosal damage and have elucidated its mechanism of action. The research findings can serve as a reference for the preparation and development of CGGE peptide products and their application in the repair of gastric mucosal damage.
